I'm with Alen, by cupping the carb you cleared some muck booking a jet, this means that although you have new tank, lines, rebuilt carb and such, there was muck in there. Where there's muck, there's more muck usually. I'd fit a filter back at the tank, blow out the fuel lines, strip and clean carb and pump(s). Then you know your clean.

I had the same thing happen when I installed a new gas tank in my 41 pickup.
Ran good for twenty or so miles, before the power loss occurred. Tried a new fuel pump, problem persisted. Sometimes the old adage "who's new in the neighborhood" comes into play. Pulled the fuel line off at the tank and fuel pump, blew air through the fuel pump end, out came a piece of thick brown string a-bought six inches long. Somehow it got into the tank before shipping. Do as ford38v8 suggested. I would also disconnect the fuel line from the carb place the end of the line in a container and crank the engine cycling the fuel pump a few squirt to eliminate any further lingering chunks of crud.
