Yes, it makes a big difference to heating up an engine. In fact I had to roll the cardboard down some on the Merc when I took it out for the first snow drive in early November. Heat gauge was higher than I saw it in the heat of July.
Cardboard up front of the grille or against the radiator was standard winter driving technique here before the fancy, store bought winter fronts were available.
